It is most important that the blender jar, glass syringe barrel, rubber coupling and filter funnels are thoroughly cleaned after each assay as residues from previous assays can produce inaccurate results. All materials and reagents must be decontaminated by soaking for at least 30 minutes in a solution of sodium hypochlorite (approx. 5% (w/w)) followed by the addition of acetone (5% by volume) for 30 minutes. Washing is then carried out as per normal practice prior to rinsing thoroughly with distilled water. It is recommended that the glass syringe barrel is rinsed through with methanol and then distilled water at the end of each assay.
Note: Aflatoxins can easily be adsorbed onto the surface of glassware resulting in reduced aflatoxin recoveries from a sample. To overcome this problem, it is recommended that tubes or vials coming into contact with solutions of aflatoxin (i.e. standards or sample eluants) should be soaked in dilute acid (eg sulphuric acid 2 mol/l) for several hours, then rinsed well with distilled water to remove all traces of acid prior to use.
